Once loaded, these booster packs are available from all of your civs, being represented by specific regions on your map, which you visit to play the quests relevant to each pack.
Not only do the booster packs contain fun quests in their own right, but they can be useful in levelling up all of your civs, either by providing useful consumables and advisors, or by allowing you access to quests which are rich in XP.
I'll go through each of these, in no partiucular order.

Skirmish
Skirmish is perhaps the more traditional form of Age of Empires, where you objective is either to conquer your opponent or build a wonder first.
The Skirmish Hall can be found on Cyprus, and there you will find a variety of options, including 22 different maps, eight different rulers, and other options.

At the start of each of your civs, if you have the Skirmish booster pack, defeating each of the eight rulers allows you to win eight different epic advisors.
You also have a number of milestones you can reach which can give you rewards, including decorations for your capital city and large cash prizes. If you reach 1,000 Skirmish victories, that's worth 50,000 coin.
At level 40, plenty of Alliance Points are there for the taking by beating unstoppable opponents.

Elite Meet at Crete
The name of this booster pack actually applies to the level 40 quest in the booster pack, but leading up to that are a series of easier challenges at level 13, 26 and 39.
They all follow a similar formula where you protect a wonder against a series of 10, 20, 30 or 40 raids by the enemy.
You travel to Crete to play the quests in this booster pack.

The rewards aren't overly generous, unless you're playing at level 40, but you can still earn alliance points and you can earn special materials to build decorations in your capital city.
One useful thing, if you happen to have a civ your building up, it can be useful winning that level 13 and level 26 gear to transfer to that civ which might be around that level.

Fertile Crescent
This booster pack comprises a series of quests which are located around Babylon, in fact, that is the region you travel to.

One thing I can say about the Fertile Crescent is that there are a lot of fun quests here, so it's worthwhile for that alone.
If you are levelling up a civ, it's useful to throw in a few Fertile Crescent quests to quicken the pace a bit.
The Ambrosia Obsession is a series of six small quests with a reward of 5,000 coin at the end of it.
I won't spoil the surprise, but the arrival of King Tut is one of the funniest moments on AoEO.

Northern Hold
You visit the region of the same name to play the Northern Hold quests.

A unique aspect about Northern Hold is the series of quests have a capture the flag type of objective (which is not my cup of team I have to admit).
The key thing I recommend about Northern Hold is that there are a few consumables which are worth winning. The best ones increase your XP from quests by 50% for a 24 hour period - you want to have that one for each of the civs you are levelling up.

All in all, the four booster packs all offer something different, and are useful in helping you level up your civs.
